Gene Name
TP53: Tumor protein p53
Gene ID
7157
General Description
This gene encodes a tumor suppressor protein containing transcriptional activation, DNA binding, and oligomerization domains. The encoded protein responds to diverse cellular stresses to regulate expression of target genes, thereby inducing cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism.

Function of the protein in oral and tooth development

Mechanistic target of rapamycin (mTOR), a highly conserved Ser/Thr protein kinase, consists of two distinct complexes: mTOR complex 1 (mTORC1) and mTOR complex 2 (mTORC2).

Luo et al, (2021) investigated the role of mTORC1 in odontoblast proliferation and mineralization. Inhibiting p53 alone or both mTORC1 and p53 with rapamycin and a p53 inhibitor, the authors found that p53 acts downstream of mTORC1; and mTORC1 promotes odontoblast mineralization. The overall findings confirm that mTORC1 upregulates odontoblast mineralization via the p53 pathway (Luo et al, 2021).
